Tadalafil is a phosphodiesterase type 5 (PDE5) inhibitor that prevents the breakdown of cyclic GMP (cGMP). Increased cGMP leads to relaxation of smooth muscle in the pulmonary arteries, penis, prostate, and bladder, causing vasodilation and improved blood flow. During sexual stimulation, nitric oxide release triggers cGMP production, which tadalafil sustains by inhibiting PDE5, thereby enhancing erectile function.
